WB (Western Blot)
(Figure 1. Western blot analysis of CDC7 using anti-CDC7 antibody (AAA126121).Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel)/90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.Lane 1: human Jurkat whole cell lysates,Lane 2: human MCF-7 whole cell lysates,Lane 3: human Hela whole cell lysates.After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-CDC7 antigen affinity purified polyclonal antibody (#AAA126121) at 0.5 ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for CDC7 at approximately 64 kDa. The expected band size for CDC7 is at 64 kDa.)

Background: Complement C2 is a protein that in humans is encoded by the C2 gene. Component C2 is a serum glycoprotein that functions as part of the classical pathway of the complement system. Activated C1 cleaves C2 into C2a and C2b. The serine proteinase C2a then combines with complement factor 4b to create the C3 or C5 convertase. Deficiency of C2 has been reported to associated with certain autoimmune diseases and SNPs in this gene have been associated with altered susceptibility to age-related macular degeneration. This gene localizes within the class III region of the MHC on the short arm of chromosome 6. Alternative splicing results in multiple transcript variants encoding distinct isoforms. Additional transcript variants have been described in publications but their full-length sequence has not been determined.
Principle of the Assay: The Picokine™ Human C2 Pre-Coated ELISA (Enzyme-Linked Immunosorbent Assay) kit is a solid-phase immunoassay specially designed to measure Human C2 with a 96-well strip plate that is pre-coated with antibody specific for C2. The detection antibody is a biotinylated antibody specific for C2. The capture antibody is monoclonal antibody from mouse and the detection antibody is biotinylated polyclonal antibody from goat. The kit contains recombinant Human C2 with immunogen: Expression system for standard: NS0; Immunogen sequence: A21-L752. The kit is analytically validated with ready-to-use reagents. To measure Human C2, add standards and samples to the wells, then add the biotinylated detection antibody. Wash the wells with PBS or TBS buffer, and add Avidin-Biotin-Peroxidase Complex (ABC-HRP). Wash away the unbounded ABC-HRP with PBS or TBS buffer and add TMB. TMB is an HRP substrate and will be catalyzed to produce a blue color product, which changes into yellow after adding the acidic stop solution. The absorbance of the yellow product at 450nm is linearly proportional to Human C2 in the sample. Read the absorbance of the yellow product in each well using a plate reader, and benchmark the sample wells' readings against the standard curve to determine the concentration of Human C2 in the sample.
