Principle of the Assay: The LumiAb Human IL-12 Chemiluminescent ELISA Kit contains the components necessary for quantitative determination of natural or recombinant Human IL-12 concentrations within any experimental sample including cell lysates, serum and plasma. This particular immunoassay utilizes the quantitative technique of a "Sandwich" Enzyme-Linked Immunosorbent Assay (ELISA) where the target protein (antigen) is bound in a "sandwich" format by the primary capture antibodies coated to each well-bottom and the secondary detection antibodies added subsequently by the investigator. The capture antibodies coated to the bottom of each well are specific for a particular epitope on Human IL-12 while the user-added detection antibodies bind to epitopes on the captured target protein. Amid each step of the procedure, a series of wash steps must be performed to ensure the elimination of non-specific binding between proteins to other proteins or to the solid phase. After incubation and "sandwiching" of the target antigen, a peroxidase enzyme is conjugated to the constant heavy chain of the secondary antibody (either covalently or via Avidin/Streptavidin-Biotin interactions), allowing for a sensitive luminescent reaction to ensue upon substrate addition. When the Peroxide Enhancer solution is added, the reaction catalyzed by peroxidase yields light that is representative of the antigen concentration. After a brief incubation, the microplate can be read with a luminometer, allowing for generation of a standard curve and subsequent determination of protein concentration!!Background/Introduction: Human IL-12 or Interleukin-12, also known as Natural Killer Cell Stimulatory Factor is composed of 2 subunits, encoded by the IL12A and IL12B genes located at loci 3q25.33-3q26 and 5q31.1-5q33.1 on chromosome 3 and 5, respectively. Composed of 2 subunits at 219 and 328 residues, respectively, the IL-12 heterodimeric cytokine can act as a growth factor for activated T-and NK-cells, enhance the lytic activity of NK/lymphokine-activated killer cells and stimulate the production of IFN-gamma by resting PBMC. The cytokine is a disulfide-linked heterodimer composed of the 35-kD subunit encoded by this gene, and a 40-kD subunit that is a member of the cytokine receptor family. This cytokine is required for the T-cell-independent induction of interferon (IFN)-gamma, and is important for the differentiation of both Th1 and Th2 cells. The responses of lymphocytes to this cytokine are mediated by the activator of transcription protein STAT4. Nitric Oxide Synthase 2A (NOS2A/NOS2) is found to be required for the signaling process of this cytokine in innate immunity. The 2nd subunit of IL-12 can associate with IL-23A to form the IL-23 interleukin, a heterodimeric cytokine which functions in innate and adaptive immunity. IL-23 may constitute with IL-17 an acute response to infection in peripheral tissues. IL-23 binds to a heterodimeric receptor complex composed of IL-12RB1 and IL23R, activates the JAK-STAT signaling cascade, stimulates memory rather than naïve T-cells and promotes the production of pro-inflammatory cytokines. Furthermore, defects in IL-12B are a cause of Mendellian susceptibility to mycobacterial disease; also known as familial disseminated atypical mycobacterial infection while genetic variations in IL-12B are a cause of susceptibility to psoriasis type 11 (PSORS11).
