Src Mab (AAA28639) was diluted 1:500 in 1% BSA/TBST and incubated Overnight at 4 degree C. After washing 3x 5 min. with TBST the blots were incubated with 1:5000 Goat anti-mouse or Goat anti-rabbit secondary antibody for 1 hr at Room temperature. IF (Immunofluorescence) (Fluorescent image of A549 cell stained with SRC Antibody. A549 cells were fixed with 4% PFA (20 min), permeabilized with Triton X-100 (0.1%, 10 min), then incubated with SRC primary antibody (1:25, 1 h at 37 degree). For secondary antibody, Alexa Fluor 488 conjugated donkey anti-mouse antibody (green) was used (1:400, 50 min at 37 degree).Cytoplasmic actin was counterstained with Alexa Fluor 555 (red) conjugated Phalloidin (7units/ml, 1 h at 37 degree).SRC immunoreactivity is localized to Cytoplasm significantly.)
